20090281025 | INHIBITION OF TUMOR GROWTH AND METASTASIS BY ATF2-DERIVED PEPTIDES - The present invention relates to novel therapies for cancer and, in particular, to therapies that are particularly suited to tumor cells resistant to other types of therapies, such as radiation therapy, chemotherapy, or a combinations thereof. The invention provides methods for identifying and implementing strategies to inhibit a transcription factor involved in promoting resistance and inhibition of apoptosis. The invention provides a compound that alters ATF2 activity, specifically amino-terminal fragments of ATF2 that retain the JNK binding domain. The invention provides methods for inhibiting tumor cell growth and for sensitizing tumor cells to apoptosis with such peptides. | 11-12-2009 |
20100168162 | SELECTIVE INHIBITORS OF AKT AND METHODS OF USING SAME - The present invention describes an improved method for screening compounds for activity in inhibiting the enzymatic activity of Akt1 protein kinase, also known as Protein Kinase B, an enzyme that is believed to play a key role in the inhibition of apoptosis and thus in the etiology of cancer and other conditions, including neurodegenerative diseases. In general, the method comprises: (1) providing a plurality of compounds suspected of having Akt1 kinase inhibitory activity; (2) modeling the docking of each of the plurality of the compounds with a target binding site derived from the crystal structure of a ternary complex involving Akt1, a nonhydrolyzable ATP analogue, and a peptide substrate derived from a physiological AKT substrate such that the protein active site is defined including those residues within a defined distance from the nonhydrolyzable ATP analogue; (3) ranking the docked compounds by goodness of fit; (4) further selecting compounds from compounds high ranked by goodness of fit in docking by using one or more screening criteria; (5) optionally, visually analyzing structures of compounds selected in step (4) to remove any compounds with improbable docking geometry; and (6) experimentally testing the selected compounds from step (4) or step (5), if step (5) is performed, to determine their inhibitory activity against Akt1 in order to select compounds with Akt1 inhibitory activity. The invention also encompasses pharmaceutical compositions including compounds whose inhibitory activity against Akt1 is discovered by the screening method, as well as methods of use of the pharmaceutical compositions to treat cancer and other conditions. | 07-01-2010 |
20120095078 | METHODS OF DIAGNOSIS AND TREATMENT OF MELANOMA - Methods for the diagnosis or prognosis of melanoma by detecting expression of ATF2 and MITF in melanocytes are provided herein. Also provided are methods of treating a melanocyte proliferative disorder with agents that modulate the transcriptional activity of ATF2 and/or MITF activity. | 04-19-2012 |
20120276122 | METHODS FOR DIAGNOSIS AND TREATMENT OF CELLULAR PROLIFERATIVE DISORDERS - Methods for the diagnosis or prognosis of cellular proliferative disorders by detecting expression of PKC in cancer cells or tumor cells are provided herein. Also provided are methods for treating a melanocyte proliferative disorder with agents that modulate the translocational activity of ATF2 and/or PKCε activity. | 11-01-2012 |

20110129844 | Use of Activating Transcription Factor-2 (ATF2) for Detecting Skin Cancer - The present invention is based on the discovery of the tumor suppressor role of activating transcription factor 2 (ATF2) in non-melanoma skin cancer development. Accordingly, the invention provides methods of diagnosing a subject as having or at risk of having skin cancer. Also provided are methods of treating skin cancer in a subject, characterizing skin cancer in a subject, and identifying agents useful for treating skin cancer in a subject. | 06-02-2011 |
20120190707 | SELECTIVE INHIBITORS OF AKT AND METHODS OF USING SAME - The present invention describes an improved method for screening compounds for activity in inhibiting the enzymatic activity of Akt1 protein kinase. In general, the method comprises: (1) providing a plurality of compounds suspected of having Akt1 kinase inhibitory activity; (2) modeling the docking of each of the plurality of the compounds with a target binding site; (3) ranking the docked compounds by goodness of fit; (4) further selecting compounds; (5) optionally, visually analyzing structures of compounds selected in step (4) to remove any compounds with improbable docking geometry; and (6) experimentally testing the selected compounds from step (4) or step (5), if step (5) is performed, to determine their inhibitory activity against Akt1.
